SCRAPING (MOUTH, PHARYNX, BUCCAL, TONGUE AND VISUALISED LESIONS)
Principle:
To provide diagnosis for malignant (Inflammatory lesion)
Procedure:
- If the lesion is moist.
- Open the bottle of fixative.
- Scrape the lesion with the tongue depressor.
- Smear the material onto a glass slide and immediately fix in 95% alcohol / spray fix.
- Stain with H&E / Pap stain.
- If the lesion is dry or has a necrotic and inflammatory surface.
- Open the bottle of fixative.
- Gently moisten and remove the necrotic debris with a non-absorbent cotton swab, which has been dipped in saline.
- Discard the swab and debris.
- Using a second non-absorbent cotton swab which has been moistured in saline gently rub the growing margins of the lesion.
- Quickly roll the swab onto a glass slide which is held over the open bottle of fixative and immediately drop the slide into the fixative.
- Stain with H&E / Pap stain.
